How is patch delivery tested?

From ingredient content to outcomes in people: how scientists study a patch, step by step.

Esta guía está disponible actualmente en inglés.

Loaded

The amount specified in the patch. A batch assay checks actual content.

Released

The amount that leaves the patch, under stated test conditions.

Crossing skin

Movement through a skin barrier, measured under stated test conditions.

In circulation

Exposure measured in a person using the specific product.

A useful outcome

A meaningful result demonstrated in a suitable human study.

Five separate questions. This is a conceptual research sequence, not measured 24/sync results. Evidence at one stage does not establish the next.

Patch science moves step by step: from the formulation, to the laboratory, to studies in people. Each method answers its own question. Here is what each one measures, and what a good report shows.

1. What is in the finished patch?

Identity and content measurements look at the ingredient and its quantity. A good report names the product or sample, batch, method, result and unit, so every figure is tied to the sample and test behind it.

2. What leaves the formulation?

A release experiment follows an ingredient out of its formulation under controlled conditions. The EMA’s medicinal patch guideline sets out drug release and skin permeation as two distinct performance tests, each with its own method. Read the 2014 guideline.

Researchers also look at what remains in a patch after use. FDA guidance on medicinal systems specifically addresses residual drug in used patches, a good example of how closely each stage of wear is studied. See the FDA’s 2011 residual-drug guidance.

3. What crosses a skin sample?

In a laboratory diffusion experiment, a skin sample separates an applied formulation from a collecting fluid. Researchers measure the substance in the fluid and in the skin over time. OECD Test Guideline 428 describes this approach, and each result is tied to its sample, formulation and experimental setup. Read the OECD method.

4. What is measured in people?

Human exposure studies are a further step. A strong report explains who took part, what they used, how samples were collected and how the product’s contribution was separated from the body’s own background levels.

5. What benefit was measured?

Outcome studies go one step further. A controlled study looks at whether a product changes a defined outcome, and the result stays attached to its tested population and conditions.

For example, Aeschbach and colleagues’ 2009 crossover experiment studied eight healthy adults using a specific 2.1 mg melatonin patch during daytime sleep. It measured both blood melatonin and sleep, a neat example of a study that tracks exposure and outcome together. Read the original melatonin study.

Use the question that matches your decision

Start with the question, then find the matching test. Note the product, the participants or skin model, the method and the measured result. Each step adds its own piece of the picture.

Science & standards sets out what goes into every 24/sync patch, and when we test the finished patch, the results go there first. You can also read what product certificates tell you.

A clear standard

The product-specific question

What does this mean for 24/sync?

Read our current evidence register to distinguish composition information, wider research and what remains unverified for our finished patches.
